Effect of Fermented Sayuksan on Activities of Antioxidant and Anticancer

Abstract
Sayuksan is four kinds of medical herbs which consist of Glycrrhizae uralensis Fischer, Paeonia lactiflora Pallas, Bupleurum falcatum Linne, and Poncirus trifoliata Rafinesqul. Sayuksan was co-cultured with Lactobacillus spp. strains and a Sacchar- omyces spp. strain, and fermented Sayuksan was extracted by ethyl acetate. As for 1,1-Diphenyl-2-picryl-hydrazyl (DPPH) and [2,2'-azinobis (3-ethylbenzothiazoline-6-sulfonic acid)] (ABTS) free radical scavenging activities and Superoxide dismutase (SOD)-like activity as antioxidant activities, the extract of fermented Sayuksan had antioxidant activities at a lower concentration than that of Sayuksan, itself. The anticancer activity of fermented Sayuksan was confirmed in HCT 116 cells (human colon cancer cells), HepG2 cells (hepatoma cells), HeLa cells (cervical cancer cells), and MDAMB- 231 cells (breast cancer cells). Fermented Sayuksan displayed anticancer activity against those four types of cancer cells at 500 ¥ìg/mL. HeLa cells showed a concentration-dependent decrease in viability after treatment with 10?500 ¥ìg/mL of the extract from fermented Sayuksan. HeLa cells whose proliferation was inhibited even at a low concentration of the extract from fermented Sayuksan were used to verify if the anticancer activity from fermented Sayuksan resulted from apoptosis. Apoptosis was detected by the poly ADP-ribose polymerase (PARP) and cleaved PARP expression patterns. The PARP protein cleaved at higher concentrations of the extract from both Sayuksan and fermented Sayuksan and induced apoptosis. Therefore, we conclude that inhibited proliferation of HeLa cells resulted from apoptosis induction by substances produced during fermentation.
